Carbonic Anhydrases: Potential targets for Cystic Fibrosis?

La fibrosis quística (FQ) es una enfermedad genética autosómica recesiva que afecta principalmente a los pulmones, originando secreciones anómalas y espesas de las glándulas exocrinas. La FQ clásica suele ocurrir por mutaciones en el gen que codifica para la síntesis de la proteína reguladora de la conductancia transmembrana de la fibrosis quística (CFTR) que actúa como un canal iónico mayormente, responsable de controlar el paso de cloruro hacia el citoplasma. La falla de la proteína CFTR, determina la acumulación de moco, creando un medio propicio para las bacterias. Recientemente se identificó que mutaciones en el gen de la Anhidrasa Carbónica (AC) se asocian con FQ. Esta forma atípica de FQ se caracteriza por presentar un fenotipo clínico muy similar, pero con ausencia de mutación en la proteína CFTR. La eliminación de las bacterias por células de FQ es ineficiente, y mayor el porcentaje de bacterias supervivientes. Recientemente se describió que las AC eucariotas podrías estar relacionadas con la acidificación lisosomal y su capacidad degradativa. La principal causa de morbi-mortalidad en pacientes con FQ es la afectación pulmonar por infecciones repetidas debido a Pseudomonas aeruginosa, causante del 95% de los fallecimientos. Una vez diseminadas por las vías respiratorias, se adaptan al medio y desarrollan resistencia a los antibióticos convencionales, tornándose imposible de erradicar. En estadíos avanzados de FQ, los cambios en la arquitectura del pulmón producen insuficiencia respiratoria y cardíaca. La prevención de la colonización crónica de P. aeruginosa evitar el deterioro de la función pulmonar y el desarrollo de resistencia antibiótica. Los tratamientos actuales tienden a mejoran los síntomas y la esperanza de vida de 35-48 años, siendo costosos, con internaciones periódicas, y necesitando en ocasiones trasplante pulmonar. El desarrollo de nuevas estrategias, como las que involucran a la AC, serian esenciales para intentar abordar estas enfermedades. AC participa del transporte de CO2 y HCO3-, y la regulación del pH, entre otros. Muchas de las 16 isoformas en mamíferos son consideradas blancos terapéuticos para tratar una variedad de enfermedades, debido a su potencial de ser inhibidas y activadas. Proponemos estudiar la participación de ACXII, ACII y ACIV en células epiteliales normales y células epiteliales de FQ (Δ,F508-CFTR), y su efecto en la predisposición a infecciones por P. aeruginosa.

Cystic fibrosis (CF) is an autosomal recessive genetic disease that mainly affects the lungs, causing abnormal and thick secretions from the exocrine glands. Classic CF usually occurs due to mutations in the gene that codes for the synthesis of the cystic fibrosis transmembrane conductance regulator (CFTR) protein, which acts mainly as an ion channel responsible for controlling the passage of chloride into the cytoplasm. CFTR protein failure determines mucus accumulation, creating a bacteria-conducive environment. Recently was identified that mutations in the Carbonic Anhydrase (CA) gene are associated with CF. Atypical CF characterizes by a similar clinical phenotype but in absence of CFTR protein mutation. Bacteria elimination in CF cells is inefficient, and the percentage of surviving bacteria is higher. Recently was describe that eukaryotic CA could be related to lysosomal acidification and its degradative capacity. Morbidity and mortality in CF is due to pulmonary impairment owed to repeated infections by Pseudomonas aeruginosa, causing 95% of deaths. Once disseminated through the respiratory tract, it adapts to the environment and develops resistance to conventional antibiotics becoming impossible to eradicate. In CF advanced stages, changes in lung architecture leads to respiratory and cardiac failure. Prevention of chronic P. aeruginosa colonization prevents lung function deterioration and antibiotic resistance development. Current treatments tend to improve symptoms and life expectancy by 35-48 years, with expensive and recurring hospitalizations sometimes requiring lung transplantation. New strategies development, such as those involving CA, would be essential to address these diseases. CA participates in CO2 and HCO3- transport, and pH regulation, among others. Of the 16-mammal isoforms, many are considered therapeutic targets in a variety of diseases due to their potential to be inhibited and activated. We propose to study the involvement of CAXII, CAII, and CAIV in normal epithelial cells and CF epithelial cells (Δ,F508-CFTR) and their effect on predisposition to P. aeruginosa infections.

Role of HSP27 in cisplatin-induced DNA damage response through ATR/CHK1 pathway in colon cancer tumor cells

El cisplatino (CDDP) es un agente alquilante genotóxico utilizado en el tratamiento de diversos carcinomas. Desafortunadamente, la quimioresistencia limita su éxito terapéutico, llevando a la recurrencia de la enfermedad. Por ello, el uso de tratamientos convencionales en combinación con terapias biológicas (inhibidores proteicos), se ha convertido en una herramienta muy prometedora. Diferentes mecanismos moleculares se han vinculado con la resistencia a CDDP, entre ellos, las proteínas de golpe de calor (Heat Shock Proteins, HSPs) y el sistema de reparación de malos apareamientos en el ADN (Mismatch Repair, MMR). La proteína de estrés HSP27, además de chaperona molecular, puede modular señales apoptóticas. Su elevada expresión en tumores se ha asociado con resistencia a CDDP. HSP27 podría conferir quimioresistencia inhibiendo mecanismos apoptóticos o incrementando la actividad de vías de reparación del ADN. En caso del sistema MMR, su deficiencia se ha asociado con resistencia al CDDP. Las proteínas MMR actuarían como sensores del daño, produciendo detención del ciclo celular y activando la vía ATR/CHK1, que conduce a la apoptosis. Sin embargo, aún no se ha establecido el rol de HSP27 en la modulación de estas vías. En base a lo expuesto proponemos como objetivo general: Determinar el rol de la proteína de golpe de calor HSP27 en la reparación del ADN mediada por el sistema MMR y su utilidad como blanco molecular en oncofarmacología. Y como Objetivos Específicos: 1) Determinar los efectos de HSP27 en la reparación del daño al ADN inducido por CDDP en células tumorales humanas MMR competentes/incompetentes, 2) Determinar las implicancias de HSP27 en la modulación de la vía de señalización de daño al ADN ATR/CHK1 inducida por CDDP, 3) Establecer la participación de HSP27 en la sensibilidad a CDDP en células tumorales MMR competentes/incompetentes. Para abordarlos emplearemos células de adenocarcinoma de colon humano HCT116+ch2/+ch3, MMR incompetentes/competentes, respectivamente y realizaremos la regulación negativa de HSP27, tratamiento con CDDP, estudios de daño y reparación del ADN, viabilidad celular, inhibición de la vía ATR/CHK1, inmunoblot, estudio de apoptosis y senescencia. La concreción del proyecto nos permitirá avanzar en el conocimiento del interactoma dinámico de HSP27 y dilucidar mecanismos moleculares que contribuyan a optimizar la sensibilidad a análogos del platino.

Cisplatin (CDDP) is a genotoxic alkylating agent widely used in the treatment of various carcinomas. Unfortunately, chemoresistance limits its therapeutic success, leading to disease recurrence. The use of conventional treatments in combination with biological therapies (protein inhibitors) has become a very promising tool in clinical oncology. Different molecular mechanisms have been linked to CDDP resistance, including Heat Shock Proteins (HSPs) and the DNA Mismatch Repair (MMR) system. The stress protein HSP27, in addition its molecular chaperone activities, can modulate apoptotic signals. High expressions of HSP27 have been associated with resistance to CDDP in tumors. HSP27 could confer chemoresistance by inhibiting apoptotic mechanisms or by increasing the activity of DNA repair pathways. Deficiency of the MMR system has also been associated with resistance to CDDP. MMR proteins act as DNA damage sensors, inducing cell cycle arrest and activating the ATR/CHK1 pathway, which leads to apoptosis. However, the role of HSP27 in the modulation of these pathways has not yet been established. Based on these data, we propose as general objective: To determine the role of the heat shock protein HSP27 in DNA repair mediated by the MMR system and its usefulness as molecular target in oncopharmacology. Specific Objectives: 1) To determine the effects of HSP27 on CDDP-induced DNA damage repair in MMR competent/incompetent human tumor cells, 2) To determine the implications of HSP27 in the modulation of the DNA damage signaling pathway ATR/CHK1 induced by CDDP, 3) To establish the participation of HSP27 in CDDP sensitivity in competent/incompetent MMR tumor cells. Human colon adenocarcinoma cells HCT116+ch2/+ch3, incompetent/competent MMR, respectively will be used. Negative regulation of HSP27, CDDP treatment, DNA damage and repair studies, cell viability, and inhibition of the ATR pathway will also be made. /CHK1, immunoblot, study of apoptosis and senescence. The completion of the project will allow us to advance our understanding of the dynamic interactome of HSP27 and elucidate molecular mechanisms that contribute to optimizing sensitivity to platinum analogs.

Micobioma y el carcinoma oral de células escamosas

Las bacterias, hongos, virus, protozoos y arqueas que residen en la cavidad oral y su genoma colectivo (microbioma oral) son componentes críticos de la salud y la enfermedad. Se ha demostrado que las bacterias comensales de la cavidad oral pueden estar asociadas con varios tipos de cáncer. Entre los cánceres orales relacionados con el microbioma oral, el carcinoma oral de células escamosas (COCE) es el más prevalente y estudiado. El estudio del microbioma habitual fúngico de la cavidad oral (micobioma habitual oral) revela que las bases de datos de genomas micóticos aún están incompletas y presentan una diversidad desconcertante. Por lo tanto, se desconoce el papel que puede desempeñar el micobioma habitual oral en la aparición de lesiones orales potencialmente malignas (LOPM) en la mucosa oral y en el COCE. A partir de estos antecedentes, pretendemos determinar el micobioma asociado a estas lesiones orales, utilizando un novedoso enfoque molecular. La combinación de PCR en tiempo real (q-PCR) y la secuenciación de alto rendimiento propuesta (NGS) para el estudio del reino fúngico permitirá determinar cuali y cuantitativamente el micobioma asociado a la cavidad oral de pacientes sanos, pacientes con LOPM y con COCE. El estudio comparativo de estos grupos permitirá caracterizar la población micótica, contribuyendo al establecimiento de patrones consistentes del micobioma asociado al COCE. Para el logro de los objetivos propuestos, se realizará un convenio de trabajo y cooperación internacional entre el Instituto de Fisiología y Genética Animal de la Academia de Ciencias de República Checa y la Facultad de Ciencias Médicas de la Universidad Nacional de Cuyo, en Mendoza, Argentina. Los investigadores de estas instituciones han demostrado una gran experticia en el estudio de ecosistemas microbianos complejos en los últimos años, con publicaciones internacionales que los respaldan.

The bacteria, fungi, viruses, protozoa, and archaea that reside in the oral cavity and their collective genome (oral microbiome) are critical components of health and disease. Commensal bacteria in the oral cavity may be associated with various types of cancer. Among oral cancers related to the oral microbiome, oral squamous cell carcinoma (OSCC) is the most prevalent and studied. The study of the normal fungal microbiome of the oral cavity (oral habitual mycobiome) reveals that the databases of fungal genomes are still incomplete and show a disconcerting diversity. Therefore, the role that oral habitual mycobiome may play in the development of potentially malignant oral disorders (PMOD) and OSCC is unknown. Hence, we will determine the mycobiome associated with these oral lesions, using a novel molecular approach. Combination of real-time PCR (q-PCR) and high-throughput sequencing (NGS) will determinate qualitative and quantitative of the mycobiome associated with the oral cavity of healthy patients, patients with PMOD and OSCC. The comparative study of these groups will allow characterizing the fungal population, contributing to the establishment of consistent patterns of the mycobiome associated with OSCC. To achieve the proposed objectives, an international cooperation and work agreement will be signed between the Institute of Physiology and Animal Genetics of the Academy of Sciences of the Czech Republic and the Faculty of Medical Sciences of the National University of Cuyo, in Mendoza, Argentina. Researchers from these institutions have demonstrated great expertise in the study of complex microbial ecosystems in recent years, with international publications to back them up.

El colesterol (col) es crucial para el metabolismo en general, sin embargo, en exceso es posible que alcance que genere efectos acumulativos o negativos al metabolismo local en diferentes órganos. En los testículos (Tes), tanto el túbulo seminífero (TS) como el intersticio utilizan col y lo gestionan mediante incorporación exógena y/o síntesis endógena. Para ello, existe un mecanismo molecular que censa la concentración de este lípido disparando una respuesta que activa o inhibe una vía regulada por la proteína de unión a elementos reguladores de esteroles, SREBP-2. Previamente describimos que la vía SREBP-2 está activa en Tes y que se altera en respuesta a la dieta grasa, provocando un aumento de col en el Tes. Los espermatozoides bajo estas condiciones incrementan el número de anomalías morfológicas debidas a una espermiogénesis defectuosa. Por otro lado observamos que la adición de aceite de oliva virgen extra (AOVE) a la dieta grasa mejora la colesterolemia y la calidad seminal / espermática. Se desconoce actualmente si los mecanismos que regulan el col intracelular se modifican fisiológicamente durante los ciclos normales del epitelio seminífero, y si existen cambios bajo dietas grasas o protectoras en las diferentes etapas. Los ciclos del epitelio seminífero son asociaciones progresivas de las células de la espermatogénesis y de Sertoli que se observan en los TS de mamíferos. Estos arreglos celulares se ordenan en estadios típicos para cada especie. Por lo tanto, nuestro objetivo es comprender el manejo de col en los distintos estadios del ciclo del epitelio seminífero en condiciones basales, hipercolesterolémicas y con AOVE.

Cholesterol (chol) is crucial for general metabolism. However, in excess, it can have cumulative or negative effects on local metabolism in different organs. In the testis (Tes), the seminiferous tubule (ST) and the interstitium use chol and manage it by exogenous incorporation and/or endogenous synthesis. For this, there is a molecular mechanism that sensitises the concentration of this lipid by triggering a response that activates or inhibits a pathway regulated by the sterol regulatory element binding protein (SREBP-2). We have previously described that the SREBP-2 pathway is active in Tes and is altered in response to a fatty diet, resulting in increased cholesterol in Tes. Under these conditions, spermatozoa show an increased number of morphological abnormalities due to defective spermiogenesis. In contrast, we observed that the addition of extra virgin olive oil (EVOO) to the fat diet improved cholesterol and sperm quality. It is currently unknown whether the mechanisms of intracellular cholesterol regulation are physiologically altered during normal seminal epithelial cycles, and whether there are changes under fatty or protective diets at different stages. Seminal epithelial cycles are progressive associations of spermatogenesis and Sertoli cells observed in the mammalian ST. These cellular arrangements are organised in species-typical stages. Therefore, we aim to understand the processing of cabbage at different stages of the seminiferous epithelial cycle under basal, hypercholesterolemic and EVOO conditions.

INTRODUCCIÓN: Los estudios de seguimiento de egresados proporcionan información confiable para calificar la formación que recibió y su influencia en su desempeño profesional e inserción laboral. La información obtenida de los egresados de forma actualizada resulta imprescindible en toda reforma curricular. La Facultad de Ciencias Médicas de la Universidad Nacional de Cuyo carece de información sobre los egresados de la carrera de medicina que permita saber si la formación de grado recibida, acorde al perfil de médico planteado actualmente en el plan de estudios es suficiente para lograr la adecuada inserción y desarrollo profesional.OBJETIVOS: El objetivo principal de este proyecto es conocer la opinión de los egresados de la carrera de medicina de la FCM-UNCuyo en relación a la formación de grado recibida y a su inserción laboral.MATERIAL Y MÉTODOS: Se desarrollará un estudio retrospectivo, descriptivo. La muestra incluirá a los egresados de las cohortes 2016 y 2017, que puedan ser contactados y deseen participar del estudio, considerando que tienen la capacidad de analizar críticamente los aspectos estudiados. Los egresados serán encuestados de forma virtual y se completará con información obtenida a través de entrevistas semiestructuradas realizadas a un subgrupo de la muestra. También se obtendrá información del mercado de trabajo. RESULTADOS ESPERADOS: al finalizar el trabajo se espera obtener información preliminar suficiente que permita proponer y analizar ajustes al plan de estudios si fueran necesarios. En coordinación con responsables de gestión académica se espera diseñar y organizar un programa de seguimiento permanente de graduados

Follow-up studies of graduates provide reliable information to qualify the training received and its influence on their professional performance and employment insertion. Updated information obtained from graduates is essential in any curricular reform. Facultad de Ciencias Médicas de la UNCuyo lacks information on the graduates of the medical career that allows knowing if the undergraduate training received, according to the physician professional profile currently proposed in the curriculum, is sufficient to achieve the adequate insertion and professional development.OBJECTIVES: The main objective of this project is to know the opinion of the graduates of the medical career of the FCM-UNCuyo in relation to the degree training received and their employment insertion.MATERIAL AND METHODS: A retrospective, descriptive study will be developed. The sample will include graduates of the 2016 and 2017 cohorts, who can be contacted and wish to participate in the study, considering that they have the ability to critically analyze the aspects studied. Graduates will be surveyed virtually and will be completed with information obtained through semi-structured interviews conducted with a subgroup of the sample. Labor market information will also be obtained.EXPECTED RESULTS: At the end of the work, it is expected that information obtained will allow adjustments to the study plan. In coordination with those responsible for academic management, it is expected to design and organize a permanent monitoring program for graduates

Según la Organización Mundial de la Salud (OMS) la enfermedad causante por el herpes simplex tipo 1 (VHS-1) es un problema mundial cada vez mayor con más de 3.700 millones de personas menores de 50 años infectadas en todo el mundo, lo que equivale a casi dos tercios de la población global (el 67%). El presente proyecto aplica herramientas científicas y tecnológicas para el diagnóstico e identificación de cepas circulantes de VHS-1 en la provincia de Mendoza para un pronto tratamiento antiviral. La tipificación de las cepas circulantes permitirá la agrupación de las mismas en diferentes grupos o clados que serán investigados en cuanto a la gravedad en las manifestaciones clínicas que ocasionan.

Infection with herpes simplex virus (HSV), known as herpes, is common globally. HSV type 1 (HSV-1) is typically transmitted by oral-to-oral contact and causes infection in or around the mouth (oral herpes), but it can also cause genital herpes. In 2016 (last available estimates), 3.7 billion people under the age of 50, or 67% of the population, had HSV-1 infection (oral or genital). The aim of this study is to identify and characterize HSV-1 isolates circulating in Mendoza province, Argentina.

La infertilidad puede ser definida como la incapacidad para concebir después de un año de relaciones sexuales sin métodos anticonceptivos. Este problema afecta al 10- 15% de las parejas a nivel mundial. Las microdeleciones del cromosoma Y (MCY) constituyen la segunda causa genética más frecuente de infertilidad masculina. MCY se producen en el locus Factor de azoospermia (AZF) en la banda q11.23. AZF se subdivide en tres regiones: AZFa, AZFb y AZFc.La Academia Europea de Andrología (EAA) y la Red Europea de Calidad de Genética Molecular (EMQN), recomiendan a la PCR como prueba gold estándar para detectar MCY. Sin embargo, la PCR evidencia deleciones sólo en una porción específica del cromosoma Y, limitandose la detección de otras variaciones de número de copias (CNVs) patogénicas. En la literatura, varios trabajos asocian estos CNVs con duplicaciones en el cromosoma Y, que llevan a fallas en la espermatogénesis. Los hallazgos inconsistentes sobre los efectos clínicos de las deleciones y duplicaciones parciales de AZF demuestran la necesidad de ampliar la investigación y de desarrollar técnicas que evalúen mejor estas variaciones genómicas.Se han estudiado en nuestro laboratorio, pacientes infértiles, azoospérmicos y/u oligozoospérmicos severos, donde no se han detectado deleciones o anomalías en el locus AZF con la prueba PCR. La empresa MRC Holland, propone la técnica de amplificación de sondas dependiente de ligación en multiplex (MLPA) para detectar deleciones y duplicaciones en el locus AZF. La puesta en marcha de esta técnica, otorga nuevas herramientas a especialistas para esclarecer la causa de la infertilidad. El objetivo de este trabajo es desarrollar el ensayo de MLPA para el diagnóstico de deleciones y duplicaciones del cromosoma Y, en el diagnóstico de infertilidad masculina.Se tomarán muestras de sangre de pacientes masculinos, con diagnóstico de infertilidad y/o azoospermia no obstructiva u oligozoospermia severa (espermatozoides en semen ≤, 15 millones/ml), derivados por especialistas. Se extraerá ADN utilizando la práctica Salting Out y, por las técnicas PCR multiplex y MLPA, se amplificará el locus AZF. Los productos obtenidos se evidenciarán en el Secuenciador Automático ABI3130.A partir de los resultados y experiencia obtenida, se pretende poner en marcha la técnica MLPA que complemente el diagnóstico de MCY y/o duplicaciones en aquellos pacientes donde no pudo ser detectado por la prueba PCR.

Infertility can be defined as the inability to conceive after one year of intercourse without contraception. This problem affects 10-15% of couples in the world. Y chromosome microdeletions (MCY) are the second most common genetic cause of male infertility. MCY are produced at the azoospermia factor (AZF) locus in band q11.23. AZF is divided into three regions: AZFa, AZFb, and AZFc.The European Academy of Andrology (EAA) and the European Molecular Genetics Quality Network (EMQN) recommend PCR as the gold standard test to detect MCY. However, PCR shows deletions only in a specific Y chromosome portion, limiting the detection of other pathogenic copy number variations (CNVs). In the literature, several works associate these CNVs with duplications in Y chromosome, which lead to failures in spermatogenesis. Inconsistent findings on clinical effects of AZF partial deletions and duplications demonstrate need more research and development of techniques that better assess these genomic variations.Infertile, azoospermic and severe oligozoospermic patients have been studied in our laboratory, where no deletions or abnormalities in the AZF locus have been detected with PCR test. MRC Holland company proposes multiplex ligation-dependent probe amplification (MLPA) technique to detect deletions and duplications in AZF locus. The implementation of this technique provides new tools to specialists to clarify infertility cause. The aim of the research is to develop the MLPA assay for the diagnosis of Y chromosome deletions and duplications in male infertility diagnostic.Blood samples will be taken from male patients diagnosed with infertility, non-obstructive azoospermia or severe oligozoospermia (sperm in semen ≤, 15 million/ml), referred by specialists. DNA will be extracted using the Salting Out practice and, AZF locus will be amplified by multiplex PCR and MLPA techniques. Products will be evidenced in Automatic Sequencer ABI3130.Based on the results and experience obtained, it is intended to implement the MLPA technique that complements MCY and duplications diagnostic in patients where it could not be detected by PCR test.

Role of PTP1B in the co-expression of B-catenin and HER2 in the cytoplasmic membrane in HER2+ breast cancer

Las pacientes con expresión del receptor para el factor de crecimiento epidérmico tipo 2 (HER2) presentan una peor evolución. Existen evidencias que sugieren vínculos entre HER2 y la señalización de B-catenina. Sin embargo, no existen resultados de otros grupos que describan una relación entre la distribución/localización de B-catenina y HER2 en función de la progresión tumoral ni de las moléculas que regulan su localización subcelular. Previamente, reportamos una correlación entre la localización de B-catenina en la membrana celular y la distribución de HER2 asociada a buen pronóstico en mujeres con cáncer de mama no tratadas con trastuzumab. El estado de fosforilación y desfosforilación de B-catenina mediado por PKD1 y PTP1B respectivamente, podría favorecer la localización en membrana de la proteína en cáncer de mama HER2+ asociado a un pronóstico favorable de la enfermedad. El desarrollo del proyecto revelará los mecanismos reguladores de la expresión génica de PTP1B y PKD1 relacionado al fenotipo tumoral. Los estudios en biopsias de pacientes HER2+ e in silico permitirán dilucidar los mecanismos moleculares que regulan la expresión PTP1B y PKD1 y su impacto sobre la localización de b-catenina en la membrana de las células tumorales HER2+, su relación con el pronóstico de la enfermedad y la respuesta a los tratamientos dirigidos. La metodología incluye estudios inmunohistoquímicos y procesaremos datos ómicos provenientes de las plataformas The Cancer Genome Atlas (TCGA) y de Metabric y los datos de microarrays provenientes de la plataforma Affymetrix SNP 6.0 array. Usaremos el lenguaje de programación R, que permite el análisis de grandes cantidades de datos, facilitando la obtención de resultados de alta calidad y relevancia biológica. Consideramos muy importante el estudio de la correlación entre B-catenina y HER2 localizados en membrana para conocer los mecanismos moleculares que favorecen este perfil. De esta manera, las pacientes con B-catenina en membrana podrían evitar la terapia anti-HER2 capaz de generarles resistencia y gastos económicos innecesarios.

Patients with receptor for epidermal growth factor type 2 (HER2) expression show a worse outcome. There is evidence to suggest links between HER2 and B-catenin signaling. However, there are no results from other groups describing a relationship between the distribution/localization of B-catenin and HER2 in function of tumor progression and the molecules that regulate its subcellular localization. Previously, we reported a correlation between the localization of B-catenin in the cell membrane and the distribution of HER2 associated with good prognosis in women with breast cancer not treated with trastuzumab. The phosphorylation/dephosphorylation states of B-catenin mediated by PKD1 and PTP1B, respectively, could support the membrane localization of the protein in HER2+ breast cancer associated with a favorable prognosis of the disease. The project will reveal the regulatory mechanisms of the gene expression of PTP1B and PKD1 related to the tumor phenotype. The studies in HER2+ patient?s biopsies and in silico will allow to elucidate the molecular mechanisms that regulate PTP1B and PKD1 expression and their impact on the localization of B-catenin in the membrane of HER2+ tumor cells, their relationship with the prognosis of the illness. The methodology includes immunohistochemistry studies. We will process omic data from The Cancer Genome Atlas (TCGA) and Metabric platforms and microarray data from the Affymetrix SNP 6.0 array platform. We will use the R programming language, which allows the analysis of large amounts of data, facilitating the achievement of high quality and biologically relevant results. We consider the study of the correlation between B-catenin and HER2 located in the membrane to be very important to know the molecular mechanisms that favor this profile. In this way, patients with membrane B-catenin could avoid anti-HER2 therapy capable of generating resistance and unnecessary economic costs.

Chlamydia trachomatis es la bacteria de transmisión sexual más frecuente en el mundo. En la mujer puede provocar cervicitis, obstrucción de las trompas de Falopio, embarazos ectópicos e infertilidad. Chlamydia es una bacteria intracelular obligada que posee un genoma muy reducido y cuya estrategia de multiplicación implica la explotación y la utilización de la maquinaria de la célula que infecta. Este estilo de vida parasitario implica la alteración de los niveles de expresión, distribución y/o función de muchas de las proteínas del hospedador. En este sentido numerosos trabajos han postulado que esta bacteria actúa como un factor de riesgo en el desarrollo de cáncer cervical. La superfamilia de las miosinas, codificadas en 39 genes y agrupadas en 12 clases diferentes, participa en numerosos procesos y se ha visto que en diferentes tipos de cáncer sus niveles de expresión se encuentran alterados. Nuestros resultados indican que C. trachomatis requiere de ciertas miosinas del hospedador. Miosina 1C (Myo1C) y miosina 6 (Myo6) son fundamentales tanto en la invasión como el desarrollo, mientras que la miosina no muscular 2 a (Myo2a) es necesaria para el desarrollo bacteriano. Postulamos que C. trachomatis podría estar afectando los niveles de expresión, distribución y/o función de dichas miosinas para asegurar su desarrollo y esto podría contribuir a la transformación celular. Por otro lado, el tratamiento de las infecciones clamidiales con los antibióticos clásicos ha demostrado ser ineficiente para erradicar la infección persistente. Teniendo en cuenta que las miosinas anteriormente mencionadas son fundamentales para su entrada y desarrollo, consideramos que el tratamiento con inhibidores específicos y reversibles para estas miosinas, como tratamiento complementario para erradicar las infecciones persistentes, podría ser una estrategia eficaz para su eliminación. En este sentido este proyecto permitirá determinar las alteraciones moleculares que provoca C. trachomatis en las miosinas de la célula que infecta y evaluar nuevos fármacos para su tratamiento.

C. trachomatis is the most common sexually transmitted bacteria in the world. It can cause cervicitis, obstruction of the fallopian tubes, ectopic pregnancies, and infertility in women. Chlamydia is an obligate intracellular bacterium that has a small genome and whose multiplication strategy involves the exploitation of cell machinery. This parasitic lifestyle involves the alteration in the expression rate, distribution, or function of many host proteins. In this sense, numerous works have postulated that this bacterium acts as a risk factor in the development of cervical cancer. The human myosin superfamily, encoded in 38 genes and grouped into 12 different classes, participates in numerous processes. It has been determined that the expression levels of myosin are altered in different types of cancer. Our results indicate that C. trachomatis requires 5 host myosins. Myosin 1C (Myo1C) and myosin 6 (Myo6) are critical for both invasion and development of Chlamydia. Non-muscle myosin 2a (Myo2a) is required for bacterial growth. We hypothesize that C. trachomatis affects the expression levels, distribution, or function of these myosins to ensure bacterial development, and this could contribute to cell transformation. On the other hand, chlamydial infections treatment with classical antibiotics is inefficient in eradicating persistent infection. Taking into account that myosins are essential for their entry and development, we consider that treatment with specific and reversible inhibitors for these myosins to eradicate persistent infections as a complementary treatment could be an effective strategy for their elimination. In this sense, this project will make it possible to determine the molecular alterations caused by C. trachomatis in the host cell myosins and to evaluate new drugs for its treatment.

La respuesta inflamatoria del hospedador a toxina Shiga (STx) y/o al lipopolisacarido (LPS) de E coli está involucrada en la fisiopatologia del Sindrome Uremico Hemolitico (SUH).El receptor Toll-like 4(TLR4),proteina que reconoce al LPS bacteriano,interviene en la respuesta inmune innata inicial. El LPS, en conjunto con CD14 y MD-2,se une en la membrana plasmatica a TLR4 e inicia una señal intracelular que conduce a respuesta inflamatoria. EL trafico desde la superficie celular yla ruta intracelular del complejo LPS/TLR4/MD-2 son esenciales para lograr el control de dicha respuesta.Una vez internalizado, el complejo es direccionado desde endosomas tempranos hacia lisosomas, enviado a endosomas de reciclaje y/oalreticuloTransGolgi.Rab7b,unapequeñaGTPasa, expresada selectivamente en monocitos y localizada en endosomas tardios, participa en la regulacion del trafico de receptores. Las señales generadas por la activacion de TLRs o induccion de estres en el RE, pueden integrarse y conducir al estimulo inflamatorio patologico.Hsp70,chaperona residente de RE,regula vias de señalizacion a traves de su interaccion con otras proteinas. CHIP, cochaperona de Hsp70, es una ubiquitina ligasa E3 que favorece la poliubiquitinacion de otras proteinas para ser degradadas en proteosomas. Estudiaremos la participacion de las vias endocitica y proteosomica sobre la regulacion temprana del complejo LPS/TLR4 y su efecto sobre la respuesta inflamatoria en monocitos estimulados con LPS. Analizaremos el efecto de LPS sobre la expresión de TLR4 en la superficie y en el interior celular, sobre la expresión de Rab7b y la producción y liberación de las citoquinas intracelulares TNFalfa e IL6. Estudiaremos el papel de Hsp70/CHIP en la via proteosomica sobre la regulacion temprana del complejo LPS/TLR4.Por último analizaremos la expresion de Rab7b, Hsp70 y CHIP, y la produccion y liberacion de citoquinas TNFaalfa, e IL6 en monocitos circulantes, obtenidos de ratones knockout TLR4-/- estimulados con LPS a fin de corroborar que Rab7b de la via endocitica y Hsp70/CHIP de la via de degradacion proteosomica estan involucradas en la regulacion de TLR4 y la consecuente respuesta inflamatoria.

The host inflammatory response to Shiga toxin (STx) and/or Ecoli lipopolysaccharide (LPS) is involved in the pathophysiology of Hemolytic Uremic Syndrome (HUS). Toll-like receptor 4 (TLR4), a protein that recognizes bacterial LPS, is involved in the initial innate immune response. LPS, in conjunction with CD14 and MD-2, binds to TLR4 on the plasma membrane and initiates an intracellular signal that leads to an inflammatory response. Traffic from the cell surface and the intracellular route of the LPS/TLR4/MD-2 complex are essential to achieve control of this response. Once internalized, the complex is routed from early endosomes to lysosomes, sent to recycling endosomes and/or the Trans Golgi reticulum. Rab7b, a small GTPase, selectively expressed on monocytes and localized in late endosomes, is involved in the regulation of receptor trafficking. The signals generated by the activation of TLRs or the induction of stress in the endoplasmic reticulum (ER) can be integrated and lead to the pathological inflammatory stimulus. Hsp70 chaparone, resident from the ER, regulates signaling pathways through its interaction with other proteins. CHIP, cochaperone of Hsp70, is an E3 ubiquitin ligase that favors the polyubiquitination of other proteins to be degraded in proteasomes. Objective: We will study the participation of the endocytic and proteosomal pathways in the early regulation of the LPS/TLR4 complex and its effect on the inflammatory response in LPS-stimulated monocytes. We will analyze the effect of LPS on the expression of TLR4 on the surface and inside the cell, the expression of Rab7b and the production and release of the intracellular cytokines TNFalfa and IL6. We will study the role of Hsp70/CHIP in the proteasomal pathway on the early regulation of the LPS/TLR4 complex. Lastly, we will analyze the expression of Rab7b, Hsp70 and CHIP, and the production and release of cytokines TNFalfa, and IL6 in circulating monocytes, obtained from TLR4-/- knockout mice and stimulated with LPS in order to corroborate that Rab7b of the endocytic pathway and that Hsp70/CHIP of the proteasome degradation pathway are involved in the regulation of TLR4 and the consequent inflammatory response.
