TEORÍA CUÁNTICA DE CAMPOS APLICADA AL ESTUDIO DE LOS EFECTOS PRODUCIDOS POR LAS FLUCTUACIONES DEL VACÍO EN MATERIALES REALES

QUANTUM FIELD THEORY APPLIED TO THE STUDY OF THE EFFECTS PRODUCED BY VACUUM FLUCTUATIONS IN REAL MATERIALS

El objetivo general de este proyecto consiste en analizar las consecuencias observables de las fluctuaciones cuánticas del vacío, en el marco de la teoría cuántica de campos. En particular, nos proponemos estudiar en detalle y de manera cuantitativa, el comportamiento de las fuerzas de Casimir en diferentes modelos teóricos que nos permitan incorporar propiedades de materiales reales y geometrías no triviales, que resulten de interés conceptual o con eventuales aplicaciones en nanotecnología. Estudiaremos casos con diferentes geometrías, considerando tanto medios conductores como dieléctricos, y con especial énfasis en la aplicación a sistemas planares. En particular, para espejos de espesor finito, estudiaremos el resultado de incluir efectos debidos a propagación (no-localidades) en la dirección normal a los espejos. También estamos interesados en el análisis del denominado efecto Casimir dinámico para materiales reales, donde nuestro interés está centrado en estudiar la producción de fotones y evaluar efectos disipativos en cavidades con paredes móviles y/o con propiedades electromagnéticas dependientes del tiempo. Nos proponemos generalizar los estudios existentes sobre el Efecto Casimir Dinámico en materiales tipo grafeno, al caso de superficies curvadas, o más generalmente, superficies cuya geometría dependa del tiempo. Para ello emplearemos descripciones que tengan en cuenta los grados de libertad microscópicos o ?materiales?, cuyo principal rol es el de imponer condiciones de contorno. Consideraremos modelos en donde la interacción entre el campo de vacío y los espejos se represente a través de la acción efectiva, en lugar de considerar el vacío sujeto a condiciones de contorno ideales. Utilizaremos un enfoque de Teoría Cuántica de Campos basado en integrales funcionales, mediante el cual hallaremos expresiones generales de las cantidades físicas relevantes para cada modelo propuesto. Verificaremos la consistencia de nuestros resultados con otros previos obtenidos mediante este y otros enfoques. Finalmente analizaremos ejemplos particulares donde puedan surgir fenómenos interesantes.

The general objective of this project consists of analyzing the observable consequences of vacuum quantum fluctuations, within the quantum field theory framework. In particular, our aim is to study, in a detailed and quantitatively manner, the behavior of Casimir forces in different theoretical models that allow us to incorporate properties of real materials and non-trivial geometries, which are of conceptual interest or have possible applications in nanotechnology. We will study cases with different geometries, considering both conductive and dielectric media, and with special emphasis on the application to planar systems. In particular, for mirrors of finite thickness, we will study the result of including effects due to propagation (non-localities) in the normal direction of the mirrors. We are also interested in the analysis of the so-called dynamic Casimir effect for real materials, where our interest is focused on studying photon production and evaluating dissipative effects in cavities with moving walls and/or time-dependent electromagnetic properties. We intend to generalize the existing studies on the Dynamic Casimir Effect in graphene-like materials, to the case of curved surfaces, or more generally, surfaces whose geometry depends on time. This can be achieved using descriptions that take into account the microscopic or material degrees of freedom, whose main role is to impose boundary conditions. Instead of considering the vacuum subject to ideal boundary conditions, we will consider models where the interaction between the vacuum field and the mirrors is represented through the effective action. We will use a Quantum Field Theory approach based on functional integrals, through which we will find general expressions for the relevant physical quantities in each proposed model. We will verify the consistency of our results with previous ones obtained through this and other approaches. Finally we will analyze particular examples where interesting phenomena can arise.

Study of the endosomal-lysosomal system in cellular models with phenotypes of neurodegenerative diseases.

Las enfermedades neurodegenerativas son causadas por la muerte progresiva de neuronas en diferentes regiones del sistema nervioso. Esta pérdida de las células nerviosas origina signos y síntomas neurológicos y neuropsicológicos característicos de cada una de ellas. La enfermedad de Parkinson (PD) es un desorden neurodegenerativo de origen desconocido, caracterizado por la pérdida de neuronas dopaminérgicas de la sustancia Nigra compacta (SNpc) por acumulación de material proteináceo en forma de inclusiones llamadas cuerpos de Lewy. Otro desorden neurodegenerativo es la enfermedad de Huntington (HD) causada por una expansión poliglutamínica anormal de huntingtina (htt), conduciendo a una acumulación de agregados de esta proteína en las células. En la HD, varios estudios han demostrado las características polifuncionales de la htt, tales como la formación de vesículas cubiertas con clatrina para la endocitosis, y la estabilidad de organelas del sistema endosomal-lisosomal. Los lisosomas son organelas que contienen hidrolasas que actúan en un ambiente acídico. Estas hidrolasas en su conjunto son responsables de la degradación, reciclaje y disponibilidad de nutrientes esencial para el mantenimiento de la homeostasis celular. Los lisosomas han sido asociados con enfermedades neurodegenerativas, cardiovasculares y cáncer. En las enfermedades neurodegenerativas se cree que la muerte celular es ocasionada por un aumento en la permeabilidad lisosomal (LMP) con la consecuente liberación de algunas proteasas (catepsinas D y B). Por otra parte se ha sugerido que la LMP es inducida por especies reactivas de oxígeno que se acumulan debido a una disfunción mitocondrial. Teniendo en cuenta que los lisosomas juegan un rol, como iniciadores o ejecutores de procesos apoptóticos relacionados con la mayor labilidad de su membrana convierten a esta organela en un potencial blanco terapéutico para el tratamiento de enfermedades. Algunos compuestos, conocidos como agentes lisosomotrópicos tienen propiedades básicas y lipofílicas que les permiten quedar secuestrados dentro de los lisosomas e incrementar su pH. Esta interferencia con la normal actividad lisosomal podría ser una herramienta útil para estudiar la influencia del sistema endosomal-lisosomal en la patogénesis de enfermedades.

Neurodegenerative diseases are caused by the progressive death of neurons in different regions of the nervous system. This loss of nerve cells causes neurological and neuropsychological signs and symptoms characteristic of each of them. Parkinson's disease (PD) is a neurodegenerative disorder of unknown origin, characterized by the loss of dopaminergic neurons in the substantia Nigra compacta (SNpc) due to the accumulation of proteinaceous material in the form of inclusions called Lewy bodies. Another neurodegenerative disorder is Huntington's disease (HD) caused by an abnormal polyglutamine expansion of huntingtin (htt), leading to an accumulation of aggregates of this protein in cells. In HD, several studies have demonstrated the polyfunctional characteristics of htt, such as the formation of clathrin-coated vesicles for endocytosis, and the stability of organelles in the endosomal-lysosomal system. Lysosomes are organelles that contain hydrolases that act in an acidic environment. These hydrolases as a whole are responsible for the degradation, recycling and availability of essential nutrients for the maintenance of cellular homeostasis. Lysosomes have been associated with neurodegenerative and cardiovascular diseases and cancer. In neurodegenerative diseases, cell death is believed to be caused by an increase in lysosomal permeability (LMP) with the consequent release of some proteases (cathepsins D and B). On the other hand, it has been suggested that PML is induced by reactive oxygen species that accumulate due to mitochondrial dysfunction. Taking into account that lysosomes play a role as initiators or executors of apoptotic processes related to the greater lability of their membrane, they make this organelle a potential therapeutic target for the treatment of diseases. Some compounds, known as lysosomotropic agents, have basic and lipophilic properties that allow them to be sequestered within lysosomes and increase their pH. This interference with normal lysosomal activity could be a useful tool to study the influence of the endosomal-lysosomal system in the pathogenesis of diseases.

Bosques nativos de tierras secas: detección mediante sensores remotos y efectos de los usos del suelo

Native dryland forests: remote sensing and land use effects

Las tierras secas ocupan un 47% de la superficie terrestre y 2.600 millones de personas residen en estas regiones, muchas de las cuales dependen de los bosques para satisfacer sus necesidades básicas. La sobreexplotación de los bosques de zonas áridas ha generado que se encuentren entre los ecosistemas más amenazados y menos estudiados del mundo. En nuestro país, son protegidos por la Ley N° 26.331 de Presupuestos Mínimos de Protección Ambiental de los Bosques Nativos. Sin embargo, Argentina se encuentra entre los 10 países con mayores tasas de deforestación del mundo. Los algarrobales de Mendoza, dominados por especies del género Prosopis, son los bosques con mayores limitaciones ambientales del país y los del sureste de la provincia están sometidos a grandes disturbios antrópicos. Los bosques pedemontanos asociados a cauces de agua dominados por chacay proveen importantes servicios ecosistémicos como protectores del suelo de cuencas hídricas y es probable que su uso irracional amenace su existencia en el corto plazo. En este contexto, los sensores remotos ofrecen grandes ventajas para monitorear el estado de los bosques. Si bien la resolución de las imágenes satelitales puede ser insuficiente para detectarlos, es muy relevante generar herramientas que permitan identificar a los bosques y a los cambios producidos por los usos del suelo. El objetivo general de este trabajo es promover el manejo sostenible de los bosques nativos de Mendoza. En este sentido, es indispensable conocer su condición y su distribución geográfica. Tomando como casos de estudio a los algarrobales de los departamentos de General Alvear y San Rafael y los bosques de chacay del departamento de Malargüe, se plantean los siguientes objetivos específicos: (1) ajustar metodologías para la detección de distintos tipos de bosque nativo de tierras secas mediante sensores remotos, (2) analizar los efectos de los usos del suelo sobre la estructura y dinámica de los bosques de algarrobo en San Rafael y Alvear, (3) conocer la estructura de los bosques de chacay en Malargüe y sus usos, y (4) desarrollar indicadores derivados de imágenes satelitales que permitan identificar las respuestas del bosque frente a distintos tipos de uso del suelo. Se espera que los resultados obtenidos constituyan aportes concretos a la planificación regional, en particular al Ordenamiento Territorial de Bosques Nativos de la provincia de Mendoza y al Plan Nacional de Restauración de Bosques Nativos.

Drylands occupy 47% of the earth's surface and 2.6 billion people reside in these regions, many of whom depend on forests for their basic needs. The overexploitation of dryland forests makes them the most threatened and least studied ecosystems in the world. In our country, they are protected by Law No. 26.331 Presupuestos Mínimos de Protección Ambiental de los Bosques Nativos. However, Argentina is among the 10 countries with the highest rates of deforestation in the world. The carob forests Mendoza, dominated by species of the genus Prosopis, have many environmental limitations. Those in the southeast of the province are subjected to great anthropic disturbances. The foothill forests associated with watercourses dominated by chacay provide important ecosystem services as protectors of the soil of watersheds and their irrational use will probably threaten them in the short term. In this context, remote sensing offers great advantages for monitoring the state of forests. Although the resolution of satellite images may be insufficient to detect them, it is very important to generate tools that enable the identification of forests and the changes produced by land use. Our general objective is to promote the sustainable management of the native forests of Mendoza. To that end, it is essential to know their condition and geographical distribution. We selected two case studies: the carob forests in the departments of General Alvear and San Rafael and the chacay forests in the department of Malargüe. Our specific objectives are: (1) adjust methodologies for the detection of different types of native forest through remote sensing, (2) analyze the effects of land use on the structure and dynamics of the carob forests in San Rafael and Alvear, (3) learn about the structure of the chacay forests in Malargüe and their uses, and (4) develop indicators derived from satellite images that enable the identification of forests responses to different types of land use. We expect to generate results that contribute to the regional planning, in particular to Mendoza Land Plan of Native Forests and to the National Plan for the Restoration of Native Forests.

Para establecer un viñedo se utilizan plantas de distintas variedades de Vitis vinifera a raíz desnuda o barbechos. Estos son producidos a partir de estacas leñosas enraizadas sobre sí mismas (plantas francas) o injertadas (plantas injertadas) sobre distintos tipos de portainjertos. Durante la producción en vivero, se utilizan auxinas sintéticas (ácido indol acético o ácido indol butírico) para promover la producción de raíces adventicias durante el proceso de encallado. Luego de este proceso, las plantas son cultivadas a campo en altas densidades donde son necesarias fertilizaciones frecuentes para promover el crecimiento vegetativo. A su vez, en esta etapa, se propagan fácilmente ciertas enfermedades como la agalla de corona (Agrobacterium tumefaciens), que puede depreciar totalmente la calidad de las plantas generando importantes pérdidas económicas. Las rizobacterias promotoras del crecimiento vegetal (PGPR, Plant Growth Promoting Rhizobacteria) son bacterias simbióticas de plantas, que producen ácido indol acético, promoviendo el crecimiento y desarrollo radical y foliar. Además, colonizan la rizósfera o la planta, y algunas excretan sustancias antibióticas o desplazan a otras bacterias, ejerciendo así control biológico. Con el propósito de reducir la utilización de productos de síntesis durante la etapa de viverización y sus efectos ambientales negativos asociados, el objetivo de este trabajo es evaluar el efecto de PGPR nativas de Cuyo sobre el enraizamiento, y el control biológico durante la producción de plantas, así como la promoción del crecimiento vegetal en V. vinifera. Para ello, se incubarán estacas de vid con dos cepas de PGPR nativas y su consorcio, y se determinarán diferentes parámetros del enraizamiento. Se evaluará la incidencia de A. tumefaciens y la promoción del crecimiento vegetal, en barbechos provenientes de estacas inoculadas. Las PGPR constituirían una herramienta sustentable para los viveros, que evitaría el uso de auxinas sintéticas, y ejercerían control biológico.

To establish a vineyard, different varieties of bare-root vines of Vitis vinifera are used. These are produced from own-rooted vines (ungrafted vines plants) or from grafted vines on different types of rootstocks. During the production on nursery, synthetic auxins (indole acetic acid or indole butyric acid) are used to promote the adventitious root production during the calluses process. Then, the plants are cultivated in a field nursery at high density where frequent fertilizations are necessary to promote the vegetative growth. At this stage, some diseases are easily propagated, such as the ?Crown gall? (Agrobacterium tumefaciens), which can depreciate the quality of the plants, generating high economics loses. Plant Growth Promoting Rhizobacteria (PGPR) are symbiotic plant rhizobacteria that produce indole acetic acid, promoting the root and foliar growth and development. In addition, they colonize the rhizosphere of the plant, and some excrete antibiotics substances or displace others bacteria, exerting biological control. In order to reduce the use of synthetic products during the nursery stage and its associated negative environmental effects, the objective of this work is evaluate the effect of native PGPR of Cuyo on the rooting and the biological control, during the plant production, such as the promotion of the plant growth in V. vinifera. To do this, woody cuttings of grapevine will be incubated whit two natives PGPR strains and their consortium, and different rooting parameters will be determined. The incidence of A. tumefaciens and the promotion of plant growth will be evaluated in bare-root vines from inoculated woody cuttings. The PGPR would constitute a sustainable tool for the nurseries, which would avoid the use of synthetic auxins, and would exercise biological control.

Contribution to the rational design of probes of electrical activity in excitable membranes through the simulation of the optical response of potential-sensitive dyes

Los colorantes sensibles a voltaje (CSV) son utilizados en el campo de la investigación científica y tienen promisoria aplicación en diagnóstico por imágenes de membranas excitables de neuronas y cardiomiocitos. Sin embargo, suaplicación está limitada por la sensibilidad y velocidad de respuesta de las moléculas conocidas. En el presente proyecto se propone realizar un estudio computacional de colorantes sensibles al potencial de membrana con características que los hacen interesantes ya sea para el desarrollo metodológico, o por un potencial alcance en el ámbito de diagnóstico por imágenes: di-3-ANEPPDHQ, sonda ampliamente utilizada en el ámbito científico y deactividad óptica documentada, indocianina verde, de potencial uso clínico ya que está aprobada por la FDA, y cuya actividad óptica en el infrarrojo cercano posibilita la iluminación transdermal, y las novedosas sondas fluorescentes VoltageFluor, cuya sensibilidad aumentada se debe al mecanismo de respuesta de transferencia electrónica fotoinducidaa través de cables moleculares. Los mecanismos de respuesta incluyen efectos configuracionales de la modulación del campo eléctrico en la interacción colorante-membrana, como también efectos electrónicos como el electrocromismo o fenómenos de transferencia electrónica fotoinducida. Este trabajo se centra en la hipótesis de que es posible elucidar las bases moleculares de la sensibilidad al campo eléctrico transmembrana de moléculas embebidas en bicapaslipídicas polarizadas, apelando a una combinación de potenciales híbridos QM/MM para calcular espectros electrónicos en fase condensada, y a las simulaciones moleculares y la mecánica estadística para obtener medidas termodinámicas y configuracionales de la interacción colorante-membrana. Esta metodología constituirá una herramienta para el desarrollo racional de sensores con respuesta óptica mejorada al campo eléctrico interfacial. El objetivo de largo alcance de este desarrollo es el de diseñar sensores con respuesta óptica mejorada al potencial de membrana.

Voltage-sensitive dyes (VSDs) are used in the field of scientific research and have a promising application in diagnostic imaging of excitable membranes of neurons and cardiomyocytes. However, its application is limited by the sensitivity and response speed of known molecules. In the present project, it is proposed to carry out a computational study of dyes sensitive to membrane potential with characteristics that make them interesting either for methodological development, or for a potential scope in the field of diagnostic imaging: di-3-ANEPPDHQ, probe widely used in science and documented optical activity, indocyannine green, of potential clinical use since it is approved by the FDA, and which optical activity in the near infrared enables transdermal illumination, and the novel VoltageFluor fluorescent probes, which increased sensitivity is due to the photoinduced electron transfer response mechanism through molecular wires. Response mechanisms include configurational effects of electric field modulation on dye-membrane interaction, as well as electronic effects such as electrochromism or photoinduced electron transfer phenomena. This work focuses on the hypothesis that it is possible to elucidate the molecular bases of the sensitivity to the transmembrane electric field of molecules embedded in polarized bilayers, appealing to a combination of QM/MM hybrid potentials to calculate electronic spectra in condensed phase, and to molecular simulations and statistical mechanics to obtain thermodynamic and configurational measurements of dye-membrane interaction. This methodology will constitute a tool for the rational development of sensors with improved optical response to the interfacial electric field. The long-range goal of this development is to design sensors with improved optical response to membrane potential.

Las actividades industriales generan contaminación a gran escala en el medio ambiente a causa de metales pesados y compuestos orgánicos tóxicos, debido a que en ocasiones los efluentes de estas industrias son descargados en aguas naturales. La industria textil consume gran cantidad de agua potable y emplean colorantes para la tinción de las fibras, esto genera la descarga de un gran volumen de aguas residuales con un alto porcentaje de compuestos coloreados y una composición química muy diversa. La presencia de colorantes en cuerpos de agua afecta los ecosistemas ya que disminuye la penetración de la luz en la superficie del agua, provocando la disminución del proceso de fotosíntesis en algas y alterando la actividad biológica por la disminución de la capacidad de oxigenación. Adicionalmente, pueden ocasionar graves efectos en la salud humana debido a sus propiedades carcinogénicas, mutagénicas y teratogénicas. Se estima que cada año a nivel global se descargan en forma continua alrededor de 280.000 toneladas de diversos colorantes textiles en sus correspondientes efluentes industriales. Considerando esta situación, es fundamental el desarrollo de tecnologías que permitan la descontaminación de dichas matrices a fin de garantizar la inocuidad de los efluentes.Se han empleado diferentes tratamientos para la remoción de estos colorantes desde soluciones acuosas, aguas naturales y/o efluentes industriales, entre ellos tratamientos biológicos, coagulación/floculación, oxidación electroquímica, filtración por membranas e intercambio iónico. Sin embargo, estas técnicas por lo general cuentan con la limitación de ser costosos. Como una alternativa prometedora, la adsorción se ha destacado en los últimos años en la reducción de concentraciones de colorantes textiles de matrices contaminadas a través de una forma de bajo costo y eficiente.En el presente plan de trabajo se propone innovar en el diseño de nuevos nanomateriales para ser empleados en la adsorción de colorantes textiles, mediante la funcionalización de GO con aminoácidos. Las propiedades de retención de estos nanomateriales se estudiarán mediante herramientas fisicoquímicas y computacionales. Los desarrollos se aplicarán a la remoción de colorantes textiles de aguas naturales y efluentes industriales, con el fin de realizar aportes concretos a la situación actual que se atraviesa mundialmente en materia de contaminación de agua con colorantes y estrategias para su eliminación o mitigación.

Industrial activities generate large-scale pollution in the environment due to heavy metals and toxic organic compounds, because sometimes the effluents of these industries are discharged into natural waters. The textile industry consumes a large amount of drinking water and uses dyes to dye the fibers, this generates the discharge of a large volume of wastewater with a high percentage of colored compounds and a very diverse chemical composition. The presence of dyes in bodies of water affects ecosystems since it decreases the penetration of light into the water surface, causing a decrease in the process of photosynthesis in algae and altering biological activity due to a decrease in oxygenation capacity. Additionally, they can cause serious effects on human health due to their carcinogenic, mutagenic and teratogenic properties. It is estimated that each year around 280,000 tons of various textile dyes are continuously discharged globally in their corresponding industrial effluents. Considering this situation, it is essential to develop technologies that allow the decontamination of said matrices in order to guarantee the innocuousness of the effluents.Different treatments have been used to remove dyes from aqueous solutions, natural waters and/or industrial effluents, including biological treatments, coagulation/flocculation, electrochemical oxidation, membrane filtration and ion exchange. However, these techniques generally have the limitation of being expensive. As a promising alternative, adsorption has been highlighted in recent years in reducing concentrations of textile dyes from contaminated matrices through a low-cost and efficient way.In this work plan, it is proposed to innovate in the design of new nanomaterials to be used in the adsorption of textile dyes, through the functionalization of GO with amino acids. The retention properties of these nanomaterials will be studied using physicochemical and computational tools. The developments will be applied to the removal of textile dyes from natural waters and industrial effluents, in order to make concrete contributions to the current situation in the world in terms of water contamination with dyes and strategies for its elimination or mitigation.

Germination of native species for restoration programs in the upper basin of Mendoza River

Uno de los mayores desafíos de los proyectos de restauración es contar con la provisión de ejemplares de plantas en condiciones de ser trasplantados, provenientes de semillas del reservorio genético local, capaces de establecerse en plantas adultas y en cantidad suficiente. El estudio de atributos germinativos de especies nativas permitiría generar información para planificar este tipo de iniciativas y garantizar un mayor número de plantas. El objetivo de este proyecto es estudiar la germinación de un conjunto de especies nativas de interés para la producción de plantines que serán utilizados para la restauración de la zona del Río Blanco, Potrerillos, en la cuenca alta del Río Mendoza. Esta zona fue afectada en 2019 por incendios que degradaron unas 15.000 ha en total, y alrededor de 8.000 ha en la mencionada cuenca. Por ello, se apunta a generar información sobre atributos germinativos de especies de interés para el proyecto participativo de restauración Sumá Nativas, con el objetivo de optimizar la producción de plantines en su vivero comunitario. Se colectarán semillas de un listado de diez especies que se ha definido en conjunto con las y los integrantes del proyecto de restauración y se realizarán experimentos de germinación en condiciones de vivero, con la aplicación de tratamientos pregerminativos en busca de optimizar el proceso germinativo con fines de viverización, es decir, lograr mayores porcentajes de germinación en menor tiempo. Se evaluarán diferentes atributos de la germinación de cada especie como el porcentaje de germinación y el tiempo inicial de germinación. Con esta propuesta, se espera generar información básica sobre la germinación de un conjunto de especies nativas de particular interés para el proyecto de Sumá Nativas, con un enfoque directo en la procedencia de las mismas desde las comunidades de contacto del área afectada por incendios en el Río Blanco.

One of the greatest challenges of restoration projects is to have supply of plant specimens in conditions to be transplanted, coming from seeds of the local genetic bank, capable of survive to upper stages and in enough amount. The study of germinative attributes of native species would allow to generate information for the planification this type of initiatives and to guarantee a greater number of plants.The objective of this project is to study the germination of a set of native species of interest for the restoration of the area of the Río Blanco, Potrerillos, in the upper basin of the Río Mendoza, affected in 2019 by fires that degraded 15,000 ha. It aims to generate information on germinative attributes of species of interest for Sumá Nativas, a participative restoration project, with the aim of optimizing the production of seedlings in its community nursery.Seeds will be collected from a list of ten species that have been defined together with the members of the restoration project and germination experiments will be carried out under nursery conditions, with the application of pre-germination treatments. Different attributes of the germination of each species will be evaluated, such as the percentage of germination and the initial time of germination.It is expected to generate basic information on the germination of a set of native species that are of particular interest for the Sumá Nativas restoration project, focusing on their origin from the contact communities of the area affected by fires in the Río Blanco, Potrerillos, Mendoza.

Los Rotavirus (RV) del grupo A son el agente causal de diarrea más frecuente y es la principal causa de mortalidad asociada a la diarrea entre las niñas y los niños menores de cinco años. Para disminuir la prevalencia de la mortalidad, la OMS recomendó la introducción universal de vacunas en todos los programas nacionales de inmunización de la región europea y las Américas entre el 2006 y el 2009. En Argentina, se incorporó la vacuna RotaTeq al Calendario Nacional de Vacunación en el año 2015. Aún así, un estudio global muy reciente ha demostrado una gran diferencia en la eficacia de la vacuna entre los países de ingresos altos y bajos, que correlaciona con la falta de acceso universal al agua limpia y los estándares más altos de higiene. Por lo tanto, nuestro objetivo general es contribuir con el conocimiento básico relacionado con los mecanismos moleculares de las células hospedadoras que emplean los RVs durante su ciclo de replicación, contribuyendo a su vez, al desarrollo de opciones terapéuticas factibles para proteger la población de niños susceptibles de ser infectados por RV.El RV es un virus con dsARN segmentado de la familia Reoviridae. Hasta la fecha, se ha demostrado que existe una fuerte interacción entre las gotas lipídicas (LDs, por sus siglas en inglés) de las células infectadas y el RV, crucial para el éxito del ciclo viral. Se ha demostrado que la infección induce la biogénesis de LDs y que los compuestos que interfieren con el metabolismo de las LDs inhiben la formación de VPs y la replicación viral. A su vez, resultados preliminares de nuestro grupo sugieren que la inducción de la biogénesis de LDs ocurre muy pronto en el ciclo de infección y que las LDs tienen un papel fundamental ya que el progreso de la infección ocurre concomitantemente con el consumo de las LDs. Así, la hipótesis del presente proyecto es que luego de la inducción en la biogénesis y acumulación de LDs, el RV induce la degradación de las LDs, que utiliza como fuente de energía para su ciclo de replicación y proponemos actividades específicas que, secuencialmente desarrolladas, permitirán conocer a nivel molecular la interacción entre el ciclo de replicación de RV y el metabolismo de las LDs en las células hospedadoras y la función de esta interacción.

Group A Rotavirus (RV) is the most common cause of diarrhea and is the leading cause of diarrhea-associated mortality among girls and boys under five years of age. To reduce the prevalence of mortality, the WHO recommended the universal introduction of vaccines in all national immunization programs in the European region and the Americas between 2006 and 2009. In Argentina, the RotaTeq vaccine was incorporated into the National Vaccination Calendar in 2015. Still, a very recent global study has shown a large difference in vaccine efficacy between high- and low-income countries, correlating with a lack of universal access to clean water and higher standards of hygiene. Therefore, our general objective is to contribute to the basic knowledge related to the molecular mechanisms of the host cells that RVs use during their replication cycle, contributing in turn, to the development of feasible therapeutic options to protect the population of susceptible children.RV is a segmented dsRNA virus of the Reoviridae family. To date, it has been shown that there is a strong interaction between the lipid droplets (LDs) of infected cells and the RV, crucial for the success of the viral cycle. It has been shown that infection induces LD biogenesis and that compounds that interfere with LD metabolism inhibit VP formation and viral replication. In turn, preliminary results from our group suggest that the induction of LD biogenesis occurs very early in the infection cycle and that LDs play a fundamental role since the progress of the infection occurs concomitantly with the consumption of LDs. Thus, the hypothesis of this project is that after the induction in the biogenesis and accumulation of LDs, the RV induces the degradation of LDs, which it uses as a source of energy for its replication cycle and we propose specific activities that, sequentially developed, will allow us to elucidate the interaction between the RV replication cycle and the metabolism of LDs in host cells and the function of this interaction.

El epidídimo de los mamíferos es un órgano dependiente de hormonas esteroidales que brinda el ambiente propicio (debido a su actividad endocítica-secretoria) al proceso de maduración progresiva de los espermatozoides que provienen del testículo. La secreción epididimaria es rica en proteínas lisosomales, un hecho llamativo, del cual aún no se conoce el mecanismo que participa en dicho proceso. En la mayoría de los tipos celulares se conocen dos tipos de receptores a manosa 6 fosfato (CD- y CI- MPR) involucrados en el transporte de las hidrolasas ácidas hacia los lisosomas. En algunos modelos celulares se ha implicado al CD-MPR como el responsable de la secreción de proteínas. Datos previos, sugieren que la enzima lisosomal catepsina D (CatD) es expresada y secretada en exceso cuando el epitelio epididimario es sometido a bajos niveles de testosterona (castración de ratas). Esta enzima puede ser transportada por los MPRs, aunque mecanismos alternativos podrían estar involucrados. Se sabe que CatD puede formar complejos con la proteína lisosomal prosaposina (Psap). Estos complejos son reconocidos por el receptor sortilina (Sor) y trasportados hacia compartimentos acídicos. Resultados obtenidos en nuestro laboratorio indican que la castración produce un aumento de los niveles del CD-MPR en el epidídimo a expensas de una disminución en los niveles de Sor. Esto sugiere la existencia de un mecanismo coordinado para el transporte de CatD que involucra a los dos tipos de receptores y el cual, a su vez, puede ser regulado por hormonas. A partir de estos antecedentes nos proponemos estudiar el modo en que las hormonas pueden activar o inactivar mecanismos coordinados de transporte (CD-MPRs y/o Sor/Psap) en el epidídimo, que conducirían a la secreción de proteínas lisosomales. El modelo propuesto incluye una línea celular epididimaria de rata (RCE-1) en donde, mediante transfección con plásmidos que inducen diferentes niveles de expresión de Sor se podrá estudiar la probable regulación estrogénica del transporte de proteínas lisosomales mediante inmunoprecipitaciones, inmunoblot e inmunofluorescencia indirecta. A su vez, en la secreción generada por estas células se podrá evaluar su incidencia sobre la maduración espermática. Incursionar en el estudio de la actividad secretoria de las células epididimarias nos ayudará a entender de qué modo se condiciona un ambiente adecuado para la maduración espermática.

The mammalian epididymis is involved in sperm maturation and its integrity and functionality depends on steroid hormones. Secretion by the epididymal epithelium is enriched in acid hydrolases, an unusual fact, since these enzymes are normally confined to lysosomes. In most cell types, the normal distribution of lysosomal proteins is mediated by mannose-6-phosphate receptors (MPRs). Two forms of these receptors have been described so far, the cation-dependent (CD-MPR) and the cation-independent (CI-MPR) MPR. CD-MPR could play a role in protein secretion. Previous data suggest that the lysosomal enzyme cathepsin D (CatD) is expressed and secreted excessively by the epididymal epithelium when testosterone levels decrease (e.g. castrated rats). CatD can be transported by the MPRs, although alternative mechanisms could be involved. It is known that CatD forms complexes with the lysosomal protein prosaposin (PSAP). In turn, these complexes are recognized by the receptor Sortilin and transported to acidic compartments. Previous results from our laboratory showed an increase of CD-MPR levels and a decrease of Sortilin due to castration, suggesting a possible interplay between both receptors and the existence of a coordinated mechanism for CatD transport involving two types of receptors and it, which could be regulated by hormones. From this, we propose to study the mechanism by which the hormones can activate or inactivate such coordinated transport mechanisms (CD-MPRs and / or Sor / Psap) in epididymis. The rat epididymal cell line RCE-1 will be used in this project. These cells will be transfect with plasmids to induce changes in Sortilin expression. The estrogenic regulation of lysosomal protein transport will be studied in these cells by immunoprecipitation, immunoblot and indirect immunofluorescence. Likewise, the effect of cell secretion on sperm maturation will be evaluated. These studies will provide new insights to understand the implicance of epididymal transport and secretion of lysosomal proteins on the sperm maturation.
