La fiebre Q (FQ), zoonosis causada por la bacteria Coxiella burnetii (Cb), se presenta como una enfermedad aguda con manifestaciones similares a la gripe hasta una neumonía severa, o crónica con endocarditis. Cb por su inusual resistencia, sobrevive en el ambiente durante semanas. Ingresa como aerosoles al organismo por vía respiratoria dispersándose a otros órganos. Afecta a trabajadores de empresas procesadoras de carnes y de zonas rurales. Se han informado casos de FQ en más de 50 países entre ellos EEUU, Francia, Alemania, España, y Holanda. En América Latina, han sido afectados Uruguay, Colombia y Brasil. En Argentina, la FQ se considera exótica, sin embargo, Cb ha sido detectada en humanos, caninos y garrapatas. Recientemente se registró un brote de FQ en un frigorífico de Entre Ríos (Comunicación Epidemiológica, Ministerio de Salud, Argentina, 08/02/2022). Ciertos patógenos ingresan a la célula hospedador quedando secuestrados en fagosomas (F) que transitan por la ruta fagocítica interactuando con endosomas hasta arribar a lisosomas, donde los patógenos suelen ser degradados. La parte final de la ruta se regula principalmente por la GTPasa Rab7 y la proteína RILP. El complejo motor dineína/dinactina asocia a los F, en forma dependiente de Rab7/RILP, con los microtúbulos sobre los cuales los F se desplazan hacia la región pericentriolar para fusionarse con endosomas tardíos y lisosomas (ET/L). Los ET/L son estaciones de tránsito del colesterol que luego se transporta hacia, por ejemplo, el retículo endoplásmico (RE), por un mecanismo que implica el contacto entre las membranas de ambos compartimientos. En este mecanismo participan proteínas transportadoras y/o detectoras de colesterol que interaccionan con Rab7/RILP. Dependiendo el patógeno que contengan, los F se fusionan con o acercan a ET/L de tal forma que el patógeno obtiene el colesterol como fuente de energía para su multiplicación intracelular. Cb se multiplica en un compartimiento llamado Vacuola que contienen Coxiella (VCC) con características de ET/L, que recluta colesterol por un mecanismo poco caracterizado. Este proyecto tiene como objetivo estudiar el rol de proteínas transportadoras y/o detectoras de colesterol en la biogénesis de la VCC. Si bien la FQ no tiene mayor incidencia en Argentina, este proyecto permitiría sumar nuevos conocimientos sobre la interacción Cb-célula hospedadora que podrían contribuir al diseño de estrategias terapéuticas para contrarrestar esta patología.

Q fever (QF), a zoonosis caused by the bacterium Coxiella burnetii (Cb), presents as an acute disease with flu-like manifestations up to severe pneumonia, or chronic with endocarditis. Due to its unusual resistance, Cb survives in the environment for weeks. It enters the body as aerosols through the respiratory tract, dispersing to other organs. It affects workers in meat processing companies and in rural areas. QF cases have been reported in more than 50 countries including the US, France, Germany, Spain, and the Netherlands. In Latin America, Uruguay, Colombia and Brazil have been affected. In Argentina, QF is considered exotic, however, Cb has been detected in humans, canines, and ticks. An outbreak of QF was recently reported in a meat processing plant in Entre Ríos (Comunicación Epidemiológica, Ministerio de Salud, Argentina, 02/08/2022). Certain pathogens enter the host cell, remaining sequestered in phagosomes (P) that travel through the phagocytic pathway, interacting with endocytic compartments until they reach lysosomes, where the pathogens are usually degraded. The final part of the pathway is regulated mainly by the GTPase Rab7 and the RILP protein. The dynein/dynactin motor complex associates P, in a Rab7/RILP-dependent manner, with microtubules on which P are transported toward pericentriolar region to fuse with late endosomes and lysosomes (ET/L). ET/L are transit stations for cholesterol, which is then transported to, for example, the endoplasmic reticulum (ER), by a mechanism that involves contact between the membranes of both compartments. Transport and/or sensor proteins of cholesterol that interact with Rab7/RILP participate in this mechanism. Depending on the pathogen they contain, the P fuse with or get close to ET/L, therefore the pathogen gets cholesterol as an energy source for its intracellular multiplication. Cb multiplies in the Coxiella-containing vacuole (CCV) with characteristics of ET/L that recruit cholesterol by a partially characterized mechanism. The aim of this project is to study the role of cholesterol transporting and/or sensing proteins in the biogenesis of CCV. Although QF lacks higher incidence in Argentina, the development of this project could add new knowledge about the Cb-host cell interaction that could contribute to the design of therapeutic strategies to counteract this pathology.

Las enfermedades cardiovasculares siguen siendo una de las principales causas de mortalidad global a pesar de las terapias basadas en la evidencia obtenida de ensayos clínicos. Si bien se asignan recursos significativos para desarrollar tratamientos más nuevos, la falta de control de la presión arterial está muy lejos de lograr objetivos adecuados. En entornos de atención médica adecuada, primaria y secundaria, la falta de un adecuado control de la presión arterial socava la terapia basada en la evidencia, lo que contribuye a cientos de miles de muertes anuales y gastos innecesarios de atención médica que superan los cientos de miles de millones de dólares en Argentina y el mundo.La iniciativa Hearts-OPS en Argentina propone diferentes estrategias para lograr este objetivo. Este proyecto rescata algunas de estas estrategias para saber si en nuestra región permiten controlar el gran flagelo de la hipertensión arterial.

Cardiovascular diseases remain one of the leading causes of global mortality despite evidence-based therapies obtained from clinical trials. While significant resources are being allocated to develop newer treatments, the lack of blood pressure control falls far short of achieving adequate goals. In adequate primary and secondary health care settings, lack of adequate blood pressure control undermines evidence-based therapy, contributing to hundreds of thousands of annual deaths and unnecessary health care expenditures in excess of hundreds of billions of dollars in Argentina and the world.The Hearts-PAHO initiative in Argentina proposes different strategies to achieve this goal. This project rescues some of these strategies to find out if they allow us to control the great scourge of arterial hypertension in our region.

ROL DE LOS FOSFOLÍPIDOS EN EXOCITOSIS ACROSOMAL COMO MOLÉCULAS DE SEÑALIZACIÓN E INDUCTORES DE CURVATURA DE MEMBRANA

PHOSPHOLIPIDS' ROLE AS SIGNALING MOLECULES AND MEMBRANE CURVATURE INDUCERS DURING ACROSOMAL EXOCYTOSIS

El espermatozoide humano exocita su gránulo secretorio (acrosoma) frente a diferentes estímulos. Este proceso se denomina reacción acrosomal (RA) y es necesario para la fertilización. La RA del espermatozoide es un modelo privilegiado para estudiar la exocitosis, aislada de otros procesos. Durante mucho tiempo se pensó que la exocitosis era mediada solamente por proteínas. Ahora está claro que la composición y la topología lipídica contribuyen a la misma conjuntamente con las proteínas. Como continuación de nuestras investigaciones pretendemos develar qué moléculas regulan la dinámica de fosfolípidos y cómo estas modifican la curvatura de membrana durante la RA. El equilibrio entre la síntesis e hidrólisis del fosfatidilinositol 4,5-bifosfato (PIP2) es fundamental para que ocurra la exocitosis. Proponemos que la concentración de este lípido regula tanto la salida de calcio de los reservorios como la curvatura de membrana. Estudiaremos ambos procesos y complementaremos la parte experimental utilizando modelos matemáticos para redes metabólicas complejas y simulando bicapas lipídicas mediante Dinámica Molecular. Proponemos: I) Establecer el mecanismo molecular por el cual se regula la síntesis de fosfatidilinositol 4,5-bifosfato (PIP2) durante la exocitosis focalizando en la fosfatidilinositol 4-fosfato 5-kinasa (PIP-5KI), II) Realizar una modelización matemática de la red metabólica y vía de señalización de fosfolípidos utilizando COPASI (COmplex PAthway SImulator), III) Establecer una correlación entre composición lipídica y curvatura de membrana en la exocitosis acrosomal.Para resolver estos interrogantes afectaremos diferentes componentes de la cascada de señalización adicionando a las gametas masculinas lípidos, proteínas recombinantes, anticuerpos, segundos mensajeros, etc. y mediremos su influencia en la exocitosis. Los resultados se analizarán a través de ensayos bioquímicos, funcionales y morfológicos. Utilizaremos TEM para monitorear la distribución de moléculas lipídicas en diferentes estadíos de la RA con sondas acopladas a nanopartículas de oro. Corroboraremos nuestros resultados, simulando ciclos de retroalimentación de la red metabólica de fosfolípidos mediante el uso del modelo matemático COPASI. Cuantificaremos la correlación entre composición lipídica y curvatura de membrana usando técnicas de biofísica computacional. Abordaremos el rol de los fosfolípidos en la RA desde un enfoque multidisciplinario.

The human sperm releases its secretory granule (acrosome) when challenged with different stimuli. This process named the acrosome reaction (AR) is necessary for fertilization. The AR of the sperm is a privileged model to study exocytosis, isolated from other processes. For a long time, it was thought that exocytosis was mediated only by proteins. It is now clear that lipid composition and topology contribute to it jointly with proteins. As a continuation of our research, we intend to reveal which molecules regulate phospholipid dynamics and how they modify membrane curvature during AR. The balance between the synthesis and hydrolysis of phosphatidylinositol 4,5-bisphosphate (PIP2) is essential for exocytosis to occur. We propose that the concentration of this lipid regulates both the calcium efflux from the reservoirs and the membrane curvature. We will study both processes, complementing the experimental part using mathematical models for complex metabolic networks and simulating lipid bilayers through Molecular Dynamics. We propose: I) To establish the molecular mechanism by which the synthesis of phosphatidylinositol 4,5-bisphosphate (PIP2) is regulated during exocytosis, focusing on phosphatidylinositol 4-phosphate 5-kinase (PIP-5KI), II) To accomplish mathematical modeling of the metabolic network and phospholipid signaling pathway using COPASI (COmplex PAthway SImulator), III) To develop a correlation between lipid composition and membrane curvature in acrosomal exocytosis. To solve these questions, we will affect different components of the signaling cascade by adding to the male gametes, lipids, recombinant proteins, antibodies, second messengers, etc. Then, we will measure their influence on exocytosis. We will analyze the results through biochemical, functional, and morphological tests. We will use TEM to monitor the distribution of lipid molecules at different stages of the AR with probes coupled to gold nanoparticles. We will corroborate our results by simulating feedback loops of the phospholipid metabolic network using the COPASI mathematical model. We will quantify the correlation between lipid composition and membrane curvature using computational biophysics techniques. We will address the role of phospholipids in RA from a multidisciplinary approach.

Inestabilidad de microsatélites en tumores sólidos como biomarcador predictivo para inmunoterapia

Microsatellite instability in solid tumors as a predictive biomarker for immunotherapy

El uso de medicina personalizada es un recurso utilizado desde hace años en Oncología para optimizar el tratamiento de pacientes afectados por un determinado tumor. Diferentes ensayos determinaron un subtipo de cáncer de colon que presentaba una respuesta duradera a bloqueadores PD-1. Estos tumores asociaban altos niveles de expresión de PD1/PD-L1 en la inmunohistoquímica (IHQ) y una alteración en los mecanismos reparadores de ADN, evidenciado con una alta Inestabilidad de Microsatélites (MSI-H). Estos resultados se extrapolaron a otros tumores sólidos. Se postuló a MSI como un marcador predictivo de tumores sólidos con buena respuesta a la inmunoterapia (anti-PD1). Esto justifica el uso emergente de MSI como biomarcador para dirigir la terapéutica en diferentes tumores.MSI tiene varias implicancias en el manejo de pacientes con tumores sólidos: puede contribuir a la pesquisa inicial de Sindrome de Lynch (SL), correlaciona con menor invasión de ganglios linfáticos y metástasis y predice una evolución favorable, tiene un rol predictivo respecto a la respuesta a inmunoterapia en distintos tipos tumorales. La FDA aprobó, en los últimos años, inmunoterapias para pacientes con distintos tipos de tumores que se basan en la inhibición de vías inmunológicas para combatir al tumor mediante el sistema inmune del paciente, siempre y cuando sean portadores de MSI-H.El proyecto plantea la utilización de MSI como biomarcador predictivo en el estudio de tumores sólidos y la incorporación del resultado de esta determinación, como guía en la decisión terapéutica oncológica. Además, la ampliación de estos estudios moleculares permitirá clasificar estos cánceres según su origen en esporádicos o hereditarios, permitiendo optimizar la evaluación y asesoramiento familiar del paciente. Objetivos: detectar MSI en tumores sólidos de pacientes posibles candidatos a tratamiento con inmunoterapia, diferenciar tumores esporádicos de hereditarios, investigar nuevos marcadores microsatélites con alta sensibilidad y especificidad para la detección de MSI y correlacionar los resultados de estudios moleculares con antecedentes clínicos y estudios anatomopatológicos y de IHQ de pacientes con sospecha de SL. Se incorporarán al estudio al menos 30 pacientes con tumores sólidos. Se estudiarán con la técnica de MSI y MS- MLPA (en casos MSI-H). Se investigarán nuevos marcadores microsatélites que permitan ampliar el reactivo de MSI desarrollado previamente por este grupo de trabajo.

The personalized medicine is a resource used for years in Oncology to optimize the treatment of patients affected by a specific tumor. Different trials have identified a subtype of colon cancer that had a long-lasting response to PD-1 blockers. These tumors associated high levels of PD1/PD-L1 expression in immunohistochemistry (IHC) and an alteration in Mis Match repair system, evidenced by high Microsatellite Instability (MSI-H). These results were extrapolated to other solid tumors. MSI has been postulated as a predictive marker of solid tumors with good response to immunotherapy (anti-PD1). This justifies the emerging use of MSI as a biomarker to target therapeutics in different tumors.MSI has several implications in the management of patients with solid tumors: it can contribute to the initial investigation of Lynch Syndrome (LS), it correlates with less lymph node invasion and metastasis and predicts a favorable evolution, and it has a predictive role regarding response to immunotherapy in different tumor types. In recent years, the FDA has approved immunotherapies for patients with different types of tumors that are based on the inhibition of immunological pathways to fight the tumor through the patient's immune system, as long as they are carriers of MSI-H.The project proposes the use of MSI as a predictive biomarker in the study of solid tumors and the incorporation of the result of this determination, as a guide in the oncological therapeutic decision. In addition, the expansion of these molecular studies will allow these cancers to be classified according to their origin as sporadic or hereditary, allowing optimization of the evaluation and patient´s family counseling.Objectives: to detect MSI in solid tumors of patients who are potential candidates for treatment with immunotherapy, to differentiate between sporadic and hereditary tumors, to investigate new microsatellite markers with high sensitivity and specificity for the detection of MSI, and to correlate the results of molecular studies with clinical history and pathological studies and IHC of patients with suspected LS.At least 30 patients with solid tumors will be included in the study. They will be studied with the MSI and MS-MLPA technique (in MSI-H cases). New microsatellite markers will be investigated to expand the MSI reagent previously developed by this working group.

Pomacea canaliculata es un gasterópodo de agua dulce autóctono de nuestro país señalado como un organismo modelo por el grado de conocimiento de su biología, la facilidad de crecimiento y reproducción en cultivo de laboratorio, por su tamaño adecuado para realizar muestreos de tejidos y por el crecimiento incesante de la información molecular (genómica, transcriptómica y proteómica). Además, esta especie tiene las cualidades para ser considerado un indicador de contaminación ambiental y se ha mostrado que responde a diferentes xenobióticos, como los metales o los disruptores endocrinos. El objetivo general de este proyecto es potenciar los recursos de los laboratorios de investigación de los Dres. Giraud-Billoud y Vega (FCM-UNCuyo) y consolidar una línea de investigación de tóxicos ambientales, fomentando la asociación entre docentes de otras dos unidades académicas, Facultades de de Odontología (Dr. Carminatti) y de Ciencias Agrarias (Dr. Albrecht). Además, se busca sumar esfuerzos en la formación de personas, tanto a nivel de grado como posgrado. Los objetivos específicos de este proyecto incluyen estudios de (1) la capacidad del simbionte que vive en la glándula digestiva para generar una memoria de exposición a diferentes metales (acumulación), su rol en la detoxificación (eliminación) y la posibilidad de usar esta simbiosis como un bioindicador ambiental; incluye la identificación de los lugares de acumulación de metales en el simbionte y en la glándula digestiva, (2) los mecanismos de defensa antioxidantes enzimáticos y las vías de activación que pone en juego esta especie expuesta a ciclos de actividad-estivación o de actividad-hibernación, así como en presencia de diferentes tóxicos ambientales (metales y disruptores endocrinos), (3) la inducción de imposex por acción de los distintos disruptores endócrinos (testosterona, androstenediona, el péptido APGWamide, TBT, TPT, 9cisRA y Roziglitazona) en hembras de este gasterópodo y los mecanismos de acción de los mismos; proponemos el estudio de imposex en hembras adultas de 4 meses de edad expuesta a distintos disruptores endócrinos, y (4) la expresión del RXR en animales juveniles de distintos tamaños (10, 15 y 20 mm) y sexo, a fin de evaluar su participación durante la organogénesis reproductiva; esta caracterización nos permitirá evaluar posteriormente la expresión del RXR en extractos de tejido del aparato copulatorio de hembras adultas expuestas a disruptores endócrinos (TBT, TPT y 9cisRA).

Pomacea canaliculata is a freshwater mollusk that has been identified as a model organism for the degree of knowledge of their biology, ease of growth and reproduction in laboratory culture, their proper size for sampling tissue and by the growth of molecular information (genomics, transcriptomics and proteomics) in the last years. In addition, this species has the qualities to be considered an indicator of environmental contamination and responds to different xenobiotics (mercury, arsenic, uranium, cadmium or tributyltin). The overall objective of this project is to enhance the resources of the research laboratories of Drs. Giraud-Billoud and Vega (FCM-UNCuyo) and consolidate a research of environmental toxins, encouraging partnerships between teachers of two academic units of Universidad Nacional de Cuyo, Facultades de Odontología (Dr. Carminatti) and Agronomía (Dr. Albrecht). Also, we look at the formation of people, both undergraduate and postgraduate level. The specific objectives of this project include studies of (1) the ability of symbiont living in the digestive gland to generate a memory of exposure to different metals (accumulation), its role in detoxification (elimination), and the possibility of using this symbiosis as an environmental bioindicator; It includes identifying the places of accumulation of metals in the symbiont and the digestive gland, (2) the mechanisms of enzymatic antioxidant defense and its activation pathways of this species exposed to cycles of activity-estivación or activity- hibernation, and in the presence of different environmental toxics (metals and endocrine disruptors), (3) induction of imposex for the action of different endocrine disruptors (testosterone, androstenedione, the APGWamide peptide, TBT, TPT, 9cisRA and Roziglitazone) in females and its action mechanisms; We propose the study of imposex in 4 month old females by exposing various endocrine disruptors, and (4) the expression of RXR in juvenile animals of different sizes (10, 15 and 20 mm) and sex, in order to evaluate their participation in reproductive organogenesis. This characterization will allow us to further evaluate the expression of RXR in tissue extracts of copulatory apparatus of adult females exposed to endocrine disruptors (TBT, TPT and 9cisRA).

Se trata de un proyecto de inducción de modificaciones bioquímicas cerebrales inducidas por drogas psicotogénicas. Se valorará el conjunto de cambios conductuales y bioquímicos producidos, su interrelación y las modificaciones de los hallazgos con distintas dosis utilizadas. Se intentará establecer correlaciones entre los efectos de las distintas dosis de bloqueantes glutmatérgicos y los hallazgos en diversos tests conductuales. Se utilizará una batería de pruebas conductuales, intentando reproducir un conjunto sindromático propio de las psicosis esquizofrénicas, tanto en sus síntomas positivos como negativos,eincluyendo los cognitivos. Fundamentalmente, el proyecto apunta al bloqueo de la transmisión glutamatérgica cerebral como herramienta de inducción de trastornos conductuales homólogos ala signosintomatología presente en las psicosis esquizofrénicas. Se parte de hallazgos previos de nuestro laboratorio, en diversas especies animales (paloma y rata). La administración será sistémica e intracerebral, en zonas y núcleos discretos previamente estudiados (Nucleus Acumbens Septi [NAS], Amígdala [Amy], Córtex Prefrontal [PFC] e Hipocampo [Hip]). Se correlacionará los hallazgos conductuales con modificaciones bioquímicas inducidas por los mismos tratamientos farmacológicos. Para tal fin, se disecará las mismas zonas cerebrales con el fin de medir modificaciones metabólicas en las mismas, inducidas por la administración sistémica de antagonistas glutamatérgicos.

This is an induction project of brain psychotogenic biochemical modifications induced by pharmacological treatment with drugs. The set of behavioral and biochemical changes produced, their interrelationship and modifications of findings with different doses used will be considered. We shall try to establish correlations between the effects of different doses of glutmatergic blockers and findings in various behavioral tests. A battery of behavioral tests will be used, trying to play a syndrome own set of schizophrenic psychoses, both positive and negative symptoms, including cognitive symptoms. Fundamentally, the project aims to blockage of cerebral glutamatergic transmission as induction tool of behavioral disorders present linked to counterparts in schizophrenic psychoses. It is part of previous findings from our laboratory, in various animal (pigeon and rat) species. The administration will be systemic and intracerebral, in areas previously studied as discrete brain nucleus (Nucleus Accumbens Septi [NAS], Amygdala [Amy], Prefrontal Cortex [PFC] and Hippocampus [Hipp]). Behavioral findings with biochemical modifications induced by these drug treatments are correlated. Aiming to this purpose, the same brain areas injected in previous studies will be desiccated and biochemically studied, in order to measure metabolic modifications induced by the systemic administration of glutamate antagonists.

La enfermedad celíaca es un trastorno autoinmune causado por la ingesta de gluten en personas genéticamente predispuestas. Su prevalencia es alta. Este estudio tiene por objeto comparar las propiedades funcionales del epitelio de celíacos con las del epitelio normal, y relacionar las variables medidas en celíacos con las características clínicas de los pacientes. En todos los casos se requerirá el consentimiento informado y se preservarán los derechos de los pacientes. En las biopsias se estudiará la corriente de cortocircuito, la resistencia eléctrica y la respuesta a secretagogos, el espesor de la capa de gel de mucus y la integridad de las uniones intercelulares. La comparación entre las propiedades in vitro de muestras de pacientes celíacos y no celíacos se realizará mediante pruebas estadísticas paramétricas o no paramétricas, según esté indicado. En los celíacos, estas variables se cotejarán con datos clínicos como edad de inicio, duración, naturaleza y severidad de los síntomas y resultados de la serología. Las variables continuas se estudiarán por regresión y las categóricas mediante comparación de proporciones. El epitelio in vitro será estudiado en cámaras de Ussing adaptadas para biopsias. La resistencia eléctrica se determinará mediante pulsos de corriente. El espesor del gel de mucus se medirá por microscopía óptica convencional y confocal. Para visualizar la estructura de las uniones intercelulares se emplearán anticuerpos marcados contra claudinas y ocludinas con el uso de microscopía confocal.

Celiac disease is an autoimmune disorder caused by the ingestion of gluten in genetically predisposed individuals. Its prevalence is high. This study aims to compare the functional properties of the epithelium of celiac disease with normal epithelium, and relate the measured variables in celiac disease with clinical features of the same patients. In all cases informed consent will be secured and patients' rights will de preserved. In biopsies short circuit current will be studied, the electrical resistance and the response to secretagogues, the thickness of the mucus gel layer and the integrity of intercellular junctions. These variables will be compared with clinical data such as age of onset, duration, nature and severity of the symptoms, and results of serology. Comparison between in vitro properties of samples of celiac and non-celiac patients will be performed by parametric or non-parametric statistical tests, as indicated. In celiac patients, continuous variables will be studied by regression and categorical variables by comparing proportions. The epithelium in vitro will be studied in Ussing chambers adapted for biopsies. The electrical resistance is determined by current pulses. The thickness of the mucus gel and be measured by conventional and confocal optical microscopy. To visualize the structure of intercellular junctions and labeled antibodies against claudins occludins using confocal microscopy will be used.

La leishmaniasis es un grupo de enfermedades parasitarias causadas por protozoarios intracelulares del género Leishmania. En Argentina, la zona endémica está comprendida por las provincias de Salta, Jujuy, Tucumán, Catamarca, Santiago del Estero, Chaco, Formosa, Misiones y Corrientes, con un registro aproximado de 300 casos nuevos anuales. Uno de los principales agentes etiológicos de amplio espectro en la presentaciones clínicas, es Leishmania (Leishmania) amazonensis, con una amplia distribución geográfica en el continente Americano, y en el Norte de Argentina. En el hospedador vertebrado la resistencia a la infección contra protozoarios del género Leishmania está dada por la inmunidad celular, en especial por la activación de los macrófagos por citoquinas sintetizadas por los linfocitos Th1. Las células del perfil Th1 secretan principalmente IL-2 e IFN-γ, mientras que las células del perfil Th2 secretan IL-4, IL-5 e IL-10. Se ha demostrado que el IFN-γ activa los macrófagos, generando la expresión de iNOS, enzima que cataliza la formación de NO, necesario para la destrucción de los amastigotes intracelulares. Por el contrario, una respuesta inmune del tipo Th2 limita la acción Th1 a través de las funciones de la IL-10 e IL-4, que desactivan a los macrófagos permitiendo el crecimiento intracelular del parasito y progresión de la enfermedad. El objetivo general del proyecto es el desarrollo y optimización de nuevas formulaciones vacunales de primera y tercera generación, basadas en antígenos totales y en antígenos recombinantes del genero Leishmania, que sean capaces de generar inmunidad humoral y celular protectora frente a la infección por L. (L.) amazonensis en un modelo murino. Por lo tanto se producirán antígenos totales de L. (L.) amazonensis (ATL) y sintetizara y purificara la proteína recombinante A2 (A2r) de L. (L.) infantum. Posteriormente se ensayaran en ratones BALB/c, protocolos de inmunización con una formulaciones vacunales basadas en ATL y A2r, en combinación con Montanide ISA 763 y Poly (I:C) como adyuvantes inductores de una respuesta inmune del tipo Th1. Finalizados los protocolos de inmunización se evaluara la respuesta inmune humoral y celular. La formulación vacunal que haya logrado una optima activación del sistema inmune, se ensayara, para evaluar la protección conferida en los animales de experimentación, contra el desafío de formas infectantes de L. (L.) amazonensis.

Leishmaniasis is a group of parasitic diseases caused by intracellular protozoa of the genus Leishmania. In Argentina, the endemic zone is composed of Salta, Jujuy, Tucumán, Catamarca, Santiago del Estero, Chaco, Formosa, Misiones and Corrientes, registering approximately 300 new cases per year. One of the main etiological agents that present a broad spectrum of clinical presentations is Leishmania (Leishmania) amazonensis, it has a wide geographical distribution in the American continent, and in the North of Argentina. Resistance against the infection with Leishmania protozoa is given by cellular immunity, especially through the activation of macrophages with citokynes synthesized by Th1 lymphocites. Th1 cells secrete mainly IL-2 and IFN-γ, while Th2 cells secrete IL-4, IL-5 and IL-10. It has been demonstrated that IFN-γ activates macrophages, generating the expression of iNOS, enzyme that catalyzes the formation of NO, necessary for the destruction of the intracellular amastigotes. On the contrary, a type Th2 immune response limits the Th1 action through the functions of IL-10 and IL-4, that deactivate the macrophages allowing the intracellular growth of the parasite and the progression of the disease. The main objective of the project is the development and optimization of a first and third generation vaccine against leishmaniasis based on total and recombinant Leishmania antigens, capable of generating humoral and cellular protective immunity against L. (L.) amazonensis infection in a murine model. Thus, total antigens (ATL) from L. (L.) amazonensis and the recombinant A2 protein from L. (L.) infantum will be produced and they will be essayed on BALB/c mice in vaccination protocols, combined with Montanide ISA 763 and Poly (I:C) as adjuvant. Once the vaccinations protocols are finished, the humoral and cellular response will be evaluated. The vaccine formulation that reaches an optimal activation of the immune system will be essayed to evaluate the given protection on animals for experimentation against the challenge of infecting forms of L. (L.) amazonensis.

Se conoce que los niveles plasmáticos de la proteína C reactiva de alta sensibilidad (PCR-us), un biomarcador inflamatorio, predicen el riesgo vascular con una estimación comparativa a la del colesterol total o lipoproteína de alta densidad. La liberación de PCR us esta mediada por interleuquina (IL) -6 y la IL-1β. Estas últimas proporciona nuevas oportunidades terapéuticas para ateroprotección. El desarrollo de este trabajo propone centrarnos en el sistema de señalización de IL-6. El presente proyecto pretende demostrar que la modulación de la respuesta inflamatoria, a través del bloqueo de los receptores de Interleuquina 6 y/o la inhibición de su cascada intracelular es uno de los principales mecanismos de la alteración vascular que provocan y mantienen el remodelado vascular. Si los resultados de este proyecto son positivos, contribuirían a confirmar la hipótesis inflamatoria de la aterogénesis y permitiría aclarar, qué rol podrían determinar las nuevas terapias basadas en la regulación de citoquinas para la prevención secundaria de enfermedades cardiovasculares, entre ellas el síndrome metabólico.

It is known that plasma levels of C-reactive protein high sensitivity (hs-CRP), an inflammatory biomarker of vascular risk predict a high estimation risk cardiovascular, comparative with total cholesterol or high density lipoprotein. The release of PCR us is mediated by interleukin (IL) -6 and IL-1β. The latter provides new therapeutic opportunities for atheroprotection. The development of this paper suggests focusing on the signaling system IL-6. his project aims to demonstrate that modulation of the inflammatory response by blocking receptor Interleukin 6 and/or inhibition of intracellular cascade is one of the main mechanisms of vascular changes that cause and maintain vascular remodeling. If the results of this project are positive, they help to confirm the inflammatory hypothesis of atherogenesis and would clarify, what role could identify new therapies based on cytokine regulation for secondary prevention of cardiovascular diseases, including metabolic syndrome.

El número y la forma normal de los espermatozoides en semen dependen de la fisiología normal del túbulo seminífero. Específicamente el epitelio seminífero genera un número de gametas masculinas en un rango típico de la especie y con una forma también especie-específica. En modelos experimentales de hipercolesterolemia adquirida se alteran ambos aspectos apareciendo en semen una oligo-terato-zoospermia. Sin embargo, una disminución de la grasaingerida asociada a ingesta de aceite de oliva mejoraron los parameros seminales alterados. En trabajos preliminares ? recientes de nuestro grupo, la eficiencia tubular se encontró disminuida y la espermiogénesis, etapa final en la formación del espermatozoide, alterada generando baja concentración seminal y cabezas anómalas asociadas o no a flagelos mal implantados en las gametas. En trabajos en marcha hemos analizado la formación de la cabeza espermática en donde el manchette (microtúbulos) que debe asociarse al anillo perinuclear, acroplaxoma y la membrana plasmática no parece distribuirse / acoplarse adecuadamente. Como consecuencia las cabezas no adquieren la forma normal. Por otro lado, en otros modelos experimentales, han observado que el incremento de colesterol en sangre se acompaña de baja tetosterona circulante. Esto podría ser un motivo de la disminución de la eficiencia tubular. En este proyecto continuaremos analizando las causas de la baja eficiencia (vinculado o no a tetosterona), las de mal formación espermáticas con énfasis en el complejo manchette- acroplaxoma-membrana celular y los efectos positivos de la adicción de aceite de oliva a la dieta.

The number and normal sperm in semen depend on the normal physiology of the seminiferous tubule. Specifically the seminiferous epithelium generates a number of male gamete in a typical range of the species and with species-specific shape also. In experimental models of hypercholesterolemia acquired by grease diet both aspects appearing as oligo-terato zoospermia. However, a decrease in the grease in diet associated to olive oil intake improved both seminal parameters altered. In preliminary work of our group, the tubular efficiency was decreased and spermiogenesis, the final stage in the formation of sperm, altered generating low concentration and abnormal heads associated or not with evil implanted of the flagellum. In ongoing work we have analyzed the formation of the sperm head where the manchette (microtubules) that must be associated with perinuclear ring, acroplaxoma and plasma membrane. But under grease diet, this special complex does not appear to be distributed / attached properly. As a result the heads do not acquire the normal way. Furthermore, in other experimental models, they have observed that increased blood cholesterol was accompanied by low circulating testosterone. This could be a reason for the decrease in tubular efficiency. This project will continue to analyze the causes of low efficiency (linked or not to tetosterona), the bad sperm formation with emphasis on complex manchette- acroplaxoma-cell membrane and the positive effects of the addition of olive oil to the diet.
